 The fully enclosed Taiaro lagoon is hypersaline (42.5 psu) and non-tidal; constant salinity and water level result from strong evaporation balanced by low percolation through the lagoon floor. Seawater can flow over the atoll rim during exceptionally high seas and may then replenish lagoonal communities with propagules of oceanic origin. The distinctive water chemistry of the lagoon suggests a possible way of identifying these immigrants. We established this potential by analysing stable isotopes of carbon and oxygen in the recent growth layers of otoliths of two adult reef fishes, Chaetodon ulietensis and Acanthurus triostegus, collected from both sides of the atoll rim. Fish from the two locations were discriminated by their isotopic signatures, suggesting that analysis of the microchemical signatures deposited during the larval development could be used in future work to determine which individuals and species complete their life-cycles in this unusual lagoon. Three DNA fragments, trs1, 2 and 3, were isolated from the Trichoderma reesei genome on the basis of their ability to promote autonomous replication of plasmids in Saccharomyces cerevisiae. Each trs element bound specifically to the isolated T. reesei nuclear matrix in vitro, and two of them bound in vivo, indicating that they are matrix attachment regions (MARs). A similar sequence previously isolated from Aspergillus nidulans (ans1) was also shown to bind specifically to the T. reesei nuclear matrix in vitro. The T. reesei MARs are AT-rich sequences containing 70%, 86% and 73% A+T over 2.9, 0.8 and 3.7 kb, respectively for trs1, 2 and 3. They exhibited no significant sequence homology, but were shown to contain a number of sequence motifs that occur frequently in many MARs identified in other eukaryotes. However, these motifs occurred as frequently in the trs elements as in randomly generated sequences with the same A+T content. trs1 and 3 were shown to be present as single copies in the T. reesei genome. The presence of the trs elements in transforming plasmids enhanced the frequency of integrative transformation of T. reesei up to five fold over plasmids without a trs. No evidence was obtained to suggest that the trs elements promoted efficient replication of plasmids in T. reseei. Retinoblastoma-binding protein 1 (RBBP1) is involved in gene regulation, epigenetic regulation, and disease processes. RBBP1 contains five domains with DNA-binding or histone-binding activities, but how RBBP1 specifically recognizes chromatin is still unknown. An AT-rich interaction domain (ARID) in RBBP1 was proposed to be the key region for DNA-binding and gene suppression. Here, we first determined the solution structure of a tandem PWWP-ARID domain mutant of RBBP1 after deletion of a long flexible acidic loop L12 in the ARID domain. NMR titration results indicated that the ARID domain interacts with DNA with no GC- or AT-rich preference. Surprisingly, we found that the loop L12 binds to the DNA-binding region of the ARID domain as a DNA mimic and inhibits DNA binding. The loop L12 can also bind weakly to the Tudor and chromobarrel domains of RBBP1, but binds more strongly to the DNA-binding region of the histone H2A-H2B heterodimer. Furthermore, both the loop L12 and DNA can enhance the binding of the chromobarrel domain to H3K4me3 and H4K20me3. Based on these results, we propose a model of chromatin recognition by RBBP1, which highlights the unexpected multiple key roles of the disordered acidic loop L12 in the specific binding of RBBP1 to chromatin.  相似文献

Geographic variation in sexual dimorphism of tooth size was assessed for the red fox Vulpes vulpes (Linnaeus, 1758) across the whole northern range of the species. Twenty-one measurements of tooth size and skull length were taken from 2849 specimens (1577 males and 1272 females) originating from 12 Nearctic and 25 Palearctic localities. The index of sexual dimorphism was calculated as a quotient of the mean measure of certain characters in males by the respective mean in females ( M m/ M f). In the whole range, the males were larger than females and mean dimorphism index of tooth size ranged from 1.01 to 1.06. On average, the tooth measurements in males were 3.6% larger than in females. The highest dimorphism was observed in the canines. Dimorphism of tooth size was higher in the Palearctic than Nearctic. Statistically significant differences between regions were found for lengths of C1, C1 and M1. In the Palearctic, higher values of the dimorphism indices were observed particularly in the southern parts of the Eurasian range of the red fox and in Great Britain. For a few metrical traits, sexual dimorphism indices presented significant relations to some geo-climatic variables. The geographic pattern of size dimorphism in the red fox seems to be shaped by sexual selection, intraspecific and interspecific competition and population density.  相似文献

The southeast Asian scarab beetle genus Peltonotus Burmeister (Scarabaeidae, Dynastinae, Cyclocephalini) is reviewed. New country records for Peltonotus morio Burmeister (Myanmar and Vietnam), Peltonotus nasutus Arrow (southern China and Cambodia), and Peltonotus favonius Jameson and Wada (Myanmar) are reported, including a new record in the Palearctic/Sino-Japanese biogeographic region. The first female specimen of Peltonotus favonius is described. Biological associations with aroid inflorescences are reviewed, and human consumption of Peltonotus beetles is reported. A key to all species, paralectotype designations for Peltonotus nasutus, diagnoses, and distributions using dynamic mapping tools are included.  相似文献

Manganese (Mn) is an essential element for humans, animals, and plants and is required for growth, development, and maintenance of health. Studies show that Mn metabolism is similar to that of iron, therefore, increased Mn levels in humans could interfere with the absorption of dietary iron leading to anemia. Also, excess exposure to Mn dust, leads to nervous system disorders similar to Parkinson's disease. Higher exposure to Mn is essentially related to industrial pollution. Thus, there is a benefit in developing a clean non-invasive technique for monitoring such increased levels of Mn in order to understand the risk of disease and development of appropriate treatments.To this end, the feasibility of Mn measurements with their minimum detection limits (MDL) has been reported earlier from the McMaster group. This work presents improvement to Mn assessment using an upgraded system and optimized times of irradiation and counting for induced gamma activity of Mn. The technique utilizes the high proton current Tandetron accelerator producing neutrons via the 7Li(p,n)7Be reaction at McMaster University and an array of nine NaI (Tl) detectors in a 4π geometry for delayed counting of gamma rays. The neutron irradiation of a set of phantoms was performed with protocols having different proton energy, current and time of irradiation. The improved MDLs estimated using the upgraded set up and constrained timings are reported as 0.67 μgMn/gCa for 2.3 MeV protons and 0.71 μgMn/gCa for 2.0 MeV protons. These are a factor of about 2.3 times better than previous measurements done at McMaster University using the in vivo set-up. Also, because of lower dose-equivalent and a relatively close MDL, the combination of: 2.0 MeV; 300 μA; 3 min protocol is recommended as compared to 2.3 MeV; 400 μA; 45 s protocol for further measurements of Mn in vivo.  相似文献

Adventitious root formation in vitro in 1-mm stem slices cut from microshoots of apple cv. Jork 9 was studied using light and electron microscopy. When indole-3-butyric acid (IBA) had been added to the medium, starch grains accumulated during the first 24 h of culture in cells of the cambial region and in cells in the vicinity of vascular tissue and in the primary rays. This accumulation occurred only in the basal part of explants. After that, the nuclei in these cells were activated, and the density of the cytoplasm and the number of cell organelles increased, whereas starch was broken down. Cambium cells started to divide transversely and at 96 h, after several divisions, a continuous ring of isodiametric cytoplasmic cells had appeared around the xylem near the basal cutting surface. The cells in this ring were rich in cell structures, and did not contain large starch grains and a central vacuole. Root meristemoids regenerated from the portions of the ring that were localized in the primary rays. From the other cells in the ring, callus developed. The meristemoids did not grow into the direction of the epidermis as in shoots, but along the vascular bundles. After emergence from the cutting surface, the meristemoids were transformed into small, dome-like primordia. They developed a typical root apex with root cap, root ground meristem and tracheid connection with shoot vascular tissue. 基于1999-2015年的MODIS NDVI时间序列遥感数据,应用趋势分析、变异系数、重标极差分析和偏相关分析等方法,分析了长江中游的植被时空变化特征及其与气象因子的关系。结果表明,长江中游地区NDVI均值总体上呈上升趋势(从0.72增加到0.80)。从空间分布来看,NDVI低值区域(0.1-0.5)占1.40%,高值区域(>0.7)占87.15%;NDVI空间格局呈"西高东低、北高南低"的分布特征,低值区域表现为以三省省会城市为中心向外辐射。Hurst指数显示,研究区大部分区域(60.54%)的NDVI变化趋势具有不确定性,持续性改善区域(34.78%)主要分布在西部山地区,持续性退化区域(3.26%)主要分布在人类活动频繁的较发达城市区域。在年际尺度上,研究区NDVI与各气象因子关系均不显著;月际尺度上,NDVI与降水、相对湿度和日照时数显著相关,降水和日照时数有明显的时滞性。区域内NDVI动态趋势以不确定性发展为主,城市群周边NDVI呈现持续退化的区域应该引起关注。  相似文献
